Multidimensional Personality Changes Following Psilocybin-Assisted Therapy in Patients With Alcohol Use Disorder: Results From a Double-Blind, Placebo-Controlled Clinical Trial

This secondary of a Phase II study (n=84) investigates the effects of psilocybin-assisted therapy (PAT) on personality traits in alcohol use disorder (AUD). Psilocybin (2x, 25-40mg/70kg; n=44) significantly reduced neuroticism and increased extraversion and openness compared to placebo (diphenhydramine, n=40). Decreased impulsiveness correlated with lower alcohol consumption post-treatment, suggesting PAT may normalize abnormal personality traits in AUD.

LSD-assisted therapy in patients with anxiety: open-label prospective 12-month follow-up

In a planned 12‑month open‑label follow‑up of a double‑blind, placebo‑controlled crossover trial, LSD‑assisted therapy produced sustained, clinically significant reductions in anxiety and depression with large effect sizes lasting up to nearly two years after treatment. Participants also showed decreased neuroticism, increased extraversion and reported positive long‑term effects attributed to the psychedelic experience.

Personality change in a trial of psilocybin therapy v. escitalopram treatment for depression

In a double‑blind randomized phase 2 trial for moderate-to-severe depression, both psilocybin therapy and escitalopram produced similar personality changes consistent with improved mental health (notably reduced neuroticism, disagreeableness and impulsivity, and increased openness), with only a possible selective increase in absorption after psilocybin. Changes following escitalopram were moderated by pre-trial positive expectancy, whereas expectancy did not moderate responses to psilocybin.
