MDMA-Assisted Therapy for Major Depressive Disorder: A Seven-Month Follow-Up Proof of Principle Trial

This long-term follow-up (n=12) of participants with moderate-to-severe major depressive disorder who received MDMA-assisted therapy (two dosing sessions integrated with nine psychotherapy sessions) found significant reductions in depression severity and functional impairment at seven months compared to baseline, with improvements maintained from the post-treatment assessment and no increases in suicidal ideation.

Investigating the safety and tolerability of single-dose psilocybin for post-traumatic stress disorder: A nonrandomized open-label clinical trial

In a Phase 2, non-randomised open-label trial of 22 adults with PTSD, a single 25 mg dose of psilocybin administered with psychological support was generally well tolerated with no serious adverse events and mostly transient side-effects, and was associated with large, clinically meaningful reductions in PTSD symptoms and improvements in functioning and quality of life sustained to 12 weeks. These results indicate single-dose psilocybin may be safe and potentially efficacious for PTSD, warranting further controlled investigation.

Psilocybin in alcohol use disorder and comorbid depressive symptoms: Results from a feasibility randomized clinical trial

In this double‑blind, randomised pilot trial of recently detoxified patients with severe alcohol use disorder and comorbid depression, two 25 mg psilocybin‑assisted psychotherapy sessions (versus 1 mg control) were feasible, acceptable and safe. At 12 weeks the 25 mg group showed higher abstinence (55% vs 11%) and significant reductions in percent drinking days and craving frequency, although blinding was imperfect and the study was small and preliminary.

MDMA-assisted therapy as a treatment for major depressive disorder: proof of principle study

In an open‑label proof‑of‑principle study of 12 participants with moderate–severe major depressive disorder, two MDMA‑assisted therapy sessions plus psychotherapy were feasible, well tolerated and produced large, statistically significant reductions in depressive symptoms (mean MADRS change −19.3) and functional impairment. No unexpected or serious adverse events occurred, and the authors conclude that randomised trials are needed to confirm efficacy.

An open-label, dose-escalation trial of psilocybin-assisted therapy for bipolar 2 depression

In an open-label, single-arm dose-escalation pilot (n=14) of psilocybin-assisted therapy for bipolar II depression, psilocybin (10 mg, with 25 mg if needed) was generally well tolerated and produced significant, sustained reductions in depressive symptoms and improved quality of life up to 90 days, with adverse events (including transient anxiety, nausea, headache and three notable psychiatric events) broadly comparable to other psilocybin studies. The authors conclude that randomised, placebo-controlled trials are needed to confirm efficacy and refine dosing protocols in this vulnerable population.

Moderating factors in psilocybin-assisted treatment affecting mood and personality: A naturalistic, open-label investigation

In a naturalistic, open‑label study a single high dose of psilocybin with psychotherapy produced sustained reductions in depression, anxiety, PTSD symptoms and neuroticism, and increases in openness and conscientiousness at three months. The size of these benefits was moderated by participants’ subjective dosing experiences (mystical‑type experiences, emotional breakthrough and post‑treatment growth) and demographic factors, highlighting variables that may help optimise psilocybin‑assisted treatment.
