Ketamine-assisted psychotherapy treatment of chronic pain and comorbid depression: a pilot study of two approaches

This observational pilot of ten people with chronic pain and comorbid major depressive disorder compared a high‑dose intramuscular "psychedelic" KAPT protocol with a low‑dose sublingual "psycholytic" protocol and found reductions in depression, pain, anxiety and PTSD symptoms overall, with larger and more consistent decreases in the psychedelic group though differences were not statistically significant given the small sample. The authors conclude KAPT may be beneficial for chronic pain/MDD comorbidity and recommend larger controlled trials to determine optimal dosing and administration.

A unified model of ketamine’s dissociative and psychedelic properties

The paper presents a unified multi-level model in which ketamine’s rapid antidepressant effects stem from acute modulation of reward circuits and a sub-acute increase in neuroplasticity, while its dissociative versus psychedelic phenomenology reflects dose- and context-dependent desegregation/disintegration of the salience network (SN) and default-mode network (DMN), respectively. Computationally, SN nodes are framed as priors about the ‘minimal’ self and DMN nodes as priors about the ‘biographical’ self, and the authors argue that relaxing high-level priors during psychedelic-assisted psychotherapy, aided by neuroplasticity, may permit revision of pathological self-models and boost long-term benefit.

Psilocin and ketamine microdosing: effects of subchronic intermittent microdoses in the elevated plus-maze in male Wistar rats

This placebo-controlled rat study (n=40) investigated the effects of ketamine (0.5-3 mg/kg) and psilocin (0.05-0.075 mg/kg) microdosing (x3) on anxiety-related explorative behaviour in rats and found that both substances caused mild anxiety as measured by a reduction of explorative behaviour on an elevated open surface.

Ketamine psychotherapy for heroin addiction: immediate effects and two-year follow-up

This randomised double-blind clinical trial (2002, n=70) found that existential psychotherapy in combination with psychedelic doses of intramuscular (im) ketamine (140mg/70kg) achieved larger results in the treatment of heroin addiction than sub-hallucinogenic doses (14mg/70kg).
