Acute effects of MDMA, MDA, lysine-MDMA, and lysine-MDA in a randomized, double-blind, placebo-controlled, crossover trial in healthy participants

In a randomized, double-blind, placebo-controlled crossover in 23 healthy volunteers, MDA produced longer‑lasting, stronger and more psychedelic‑like subjective and autonomic effects and more adverse reactions than equimolar MDMA. Lys‑MDA acted as a functional slow‑release prodrug that delayed onset and peak effects, whereas Lys‑MDMA did not release MDMA and produced no measurable effects, showing lysine conjugation can alter timing but not necessarily improve tolerability.

Treatment approaches and efficacy in Psychedelic-Induced Psychosis: A systematic review

This systematic review (n=93 cases) found that psychedelic-induced psychosis, primarily caused by LSD and MDMA, lasted an average of 1.8 weeks and responded much better to second-generation antipsychotics (91% response rate) than first-generation antipsychotics (27% response rate), though one-third of patients later developed schizophrenia spectrum disorders.

MDMA pharmacokinetics: A population and physiologically based pharmacokinetics model-informed analysis

Using clinical, published and in vitro data, the authors developed and verified population and physiologically based pharmacokinetic models for MDMA which show that a high‑fat meal delays Tmax without changing overall exposure and that split dosing (2 h apart) lowers early AUC and delays Tmax compared with a single dose. The models further indicate MDMA is a potent CYP2D6 inhibitor but is unlikely to cause clinically meaningful drug–drug interactions via renal transporters, supporting model‑informed predictions of clinically relevant dosing regimens.

This focus group study (n=19) investigated the perspectives of adolescents, parents, and clinicians on MDMA-assisted psychotherapy for adolescents with PTSD. Initial attitudes towards MDMA were mainly unfavourable, but after an explanation of the therapy, all but one participant supported its potential use, emphasizing the importance of research.

Perspectives on the therapeutic potential of MDMA: A nation-wide exploratory survey among substance users

In a nation-wide survey of 918 people with alcohol or substance use disorders, most respondents supported medical research into MDMA, believed MDMA-assisted therapy might be useful, and 58.8% were willing to try it; support and belief did not differ by race or ethnicity, though small disparities arose in willingness and concerns. These findings indicate broadly positive patient-level attitudes toward MDMA-assisted therapy and point to areas to address for equitable access and more diverse research participation.
