Ketamine induces multiple individually distinct whole-brain functional connectivity signatures

This single-blind placebo-controlled study (n=40) investigated the neural and behavioral effects of acute ketamine in healthy participants. Results revealed robust inter-individual variability in both neural and behavioral responses to ketamine, with data-driven individual symptom variation mapping onto distinct neural gradients. These findings emphasize the need to consider individual variation in response to ketamine and suggest potential implications for developing precise pharmacological biomarkers in psychiatry.

Transcriptomics-informed large-scale cortical model captures topography of pharmacological neuroimaging effects of LSD

Self-blinding citizen science to explore psychedelic microdosing

Using a self‑blinding citizen‑science protocol in the largest placebo‑controlled microdosing study to date (n=191), the authors found psychological outcomes improved after four weeks but with no significant differences between microdose and placebo groups. Small acute and anxiety effects appeared due to participants breaking blind, suggesting reported benefits of microdosing are largely placebo‑driven and demonstrating the feasibility of a low‑cost self‑blinding approach.

DMT alters cortical travelling waves

DMT increases forward (bottom‑up) cortical travelling waves and suppresses top‑down alpha rhythms, producing spatio‑temporal activation similar to visual stimulation despite eyes being closed. These effects support a predictive‑processing account in which psychedelics reduce the precision‑weighting of priors, shifting the balance toward bottom‑up signalling.

Changes in global and thalamic brain connectivity in LSD-induced altered states of consciousness are attributable to the 5-HT2A receptor

In a double‑blind, placebo‑controlled crossover in 24 healthy adults, LSD reduced associative but increased sensory‑somatomotor and thalamic global functional connectivity. These subjective and neural effects were fully blocked by the 5‑HT2A antagonist ketanserin and spatially matched cortical 5‑HT2A gene expression, implicating the 5‑HT2A receptor as the primary mediator of LSD’s brain connectivity changes.
