5-Methoxy-N,N-dimethyltryptamine (5-MeO-DMT) for alcohol use disorder: An open-label, phase 2, proof-of-concept, clinical trial

In an open‑label Phase IIa trial (n-12), a single 10 mg intranasal dose of 5‑MeO‑DMT (BPL‑003) given alongside a 10‑week relapse‑prevention CBT programme was acceptable in safety and tolerability in people with moderate–severe alcohol use disorder. Over 12 weeks, participants showed large improvements in drinking (mean abstinent days rose from 33.2% to 80.8% and heavy drinking days fell from 56.2% to 13.2%), providing preliminary evidence of efficacy and supporting larger controlled trials.

Psychedelic treatment for co-occurring alcohol misuse and post-traumatic stress symptoms among United States Special Operations Forces Veterans

In a retrospective survey of 27 US Special Operations Forces veterans who completed a psychedelic treatment programme in Mexico, self‑reported alcohol misuse and post‑traumatic stress symptoms showed marked reductions with very large effect sizes (d≈–2.4 to –2.8) and 85% reduced drinking to non‑risky levels, while psychological flexibility increased. The uncontrolled, retrospective design limits causal inference, and the authors call for rigorous longitudinal trials to determine whether psychedelic‑assisted therapy is effective in this population.

Cross-sectional associations between lifetime use of psychedelic drugs and psychometric measures during the COVID-19 confinement: A transcultural study

In a cross‑sectional online survey of 2,974 English, Portuguese and Spanish speakers during the first COVID‑19 lockdown, regular lifetime users of psychedelic drugs reported lower psychological distress and peritraumatic stress and greater perceived social support than occasional users and non‑users. They also scored higher on novelty‑seeking and self‑transcendence and lower on cooperativeness, suggesting either a protective effect of psychedelics or pre‑existing personality differences that predispose to regular use.

The serotonin hallucinogen 5-MeO-DMT alters cortico-thalamic activity in freely moving mice: Regionally-selective involvement of 5-HT1A and 5-HT2A receptors

This rodent study (2017) suggests that the hallucinatory effects of 5-MeO-DMT may be due to simultaneous alteration of prefrontal and visual brain activities. The authors point to 5-HT1A receptor antagonists as a potential treatment for visual hallucinations.
