Enhancing mindfulness and compassion through an ayahuasca-inspired formulation containing N,N-DMT and harmine: A randomized controlled trial in healthy subjects

In a randomised, double‑blind, placebo‑controlled within‑subjects trial in 31 healthy volunteers, an ayahuasca‑inspired N,N‑DMT plus harmine formulation produced acute increases in mindfulness and both self‑ and other‑directed compassion one day after dosing compared with harmine or placebo, with larger effects in high‑sensitivity individuals. These findings suggest the formulation may have therapeutic potential similar to traditional ayahuasca and warrant further clinical investigation.

Effects of a Single Dose of Ayahuasca in College Students With Harmful Alcohol Use: A Single-blind, Feasibility, Proof-of-Concept Trial

In a single-blind proof-of-concept trial in 11 college students with harmful alcohol use, one dose of ayahuasca with psychological support was well tolerated and produced a short-term reduction in days drinking at 2–3 weeks that did not survive correction for multiple comparisons. The study demonstrates feasibility of the protocol and supports larger, controlled trials to assess efficacy.

Interactive Effects of Ayahuasca and Cannabidiol in Social Cognition in Healthy Volunteers: A Pilot, Proof-of-Concept, Feasibility, Randomized-Controlled Trial

In a small randomised, placebo-controlled pilot trial in healthy volunteers, 600 mg oral cannabidiol given before ayahuasca produced no detectable interaction on facial emotion recognition, empathy tasks or subjective effects — both groups showed reduced reaction times and decreases in anxiety/sedation with no between-group differences. Ayahuasca with or without CBD was generally well tolerated, supporting feasibility of further, larger trials to confirm these null-interaction findings.

Evaluating the Potential Use of Serotonergic Psychedelics in Autism Spectrum Disorder

This review evaluates evidence that serotonergic psychedelics (5‑HT2A agonists such as LSD, psilocybin and DMT) may ameliorate social deficits and co‑occurring anxiety and depression in autism spectrum disorder. It highlights neurobiological constraints (synaptic, serotonergic, prefrontal and thalamocortical dysfunction), mixed and sometimes adverse outcomes in historical paediatric trials, and concludes that rigorous contemporary studies are needed to determine whether benefits outweigh risks and whether 5‑HT2A is a viable therapeutic target.

Effects of Ayahuasca on the Recognition of Facial Expressions of Emotions in Naive Healthy Volunteers: A Pilot, Proof-of-Concept, Randomized Controlled Trial

In a pilot randomised controlled trial of 22 healthy volunteers, a single dose of ayahuasca produced no significant acute or prolonged change in recognition of facial expressions of emotion compared with placebo. The brew was generally well tolerated but showed time-dependent alkaloid degradation (notably DMT), which—alongside dosing, learning effects and sample characteristics—may account for the negative finding and indicates future trials should ensure alkaloid stability.

A placebo-controlled study of the effects of ayahuasca, set and setting on mental health of participants in ayahuasca group retreats

In a placebo-controlled naturalistic study of 30 ayahuasca retreat participants, symptoms of depression, anxiety and stress fell after ceremonies in both ayahuasca (n=14) and placebo (n=16) groups—indicating strong set-and-setting/placebo effects—while ayahuasca specifically increased implicit emotional empathy to negative stimuli. The findings emphasise the need for placebo-controlled designs and investigation of non-pharmacological contributors in psychedelic research.
