Pharmacological effects of methylone and MDMA in humans

In a randomised, double‑blind, placebo‑controlled crossover trial in 17 experienced psychostimulant users, a single oral dose of methylone (200 mg) produced sympathomimetic effects and stimulant/entactogenic subjective effects similar to MDMA (100 mg) but with a faster onset and shorter duration. These results indicate methylone’s acute pharmacological profile and abuse potential in humans are comparable to those of MDMA.

Examining associations between MDMA/ecstasy and classic psychedelic use and impairments in social functioning in a U.S. adult sample

In a nationally representative US sample (N = 214,505), lifetime MDMA/ecstasy use was associated with lower odds of three of four past‑year social‑functioning impairments (difficulty dealing with strangers, difficulty participating in social activities, and being prevented from participating), and lifetime mescaline use was associated with lower odds of difficulty dealing with strangers, whereas other substances showed no protective association or higher odds. These cross‑sectional findings cannot establish causality and indicate the need for experimental studies to test whether MDMA or mescaline can causally improve social functioning.

Mescaline: The forgotten psychedelic

This review (2022) explores mescaline's pharmacological and behavioural effects using preclinical and clinical research. The pharmacological mechanisms of mescaline are similar to those of other classical psychedelics i.e. binding at the 5HT2A receptor. Entactogens: How the Name for a Novel Class of Psychoactive Agents Originated

The paper explains the coining of the term "entactogen" for MDMA-like drugs, arguing they are distinct from classical hallucinogenic phenethylamines by three structural features (an N‑methylated nitrogen, a reversal of stereochemical potency, and tolerance of an alpha‑ethyl substitution) and by a unique psychopharmacology characterised by prosocial, anxiolytic and introspective effects. It further summarises the established mechanism for MDMA as uptake via the neuronal serotonin transporter followed by carrier‑mediated release of stored serotonin.

Cross-sectional associations between lifetime use of psychedelic drugs and psychometric measures during the COVID-19 confinement: A transcultural study

In a cross‑sectional online survey of 2,974 English, Portuguese and Spanish speakers during the first COVID‑19 lockdown, regular lifetime users of psychedelic drugs reported lower psychological distress and peritraumatic stress and greater perceived social support than occasional users and non‑users. They also scored higher on novelty‑seeking and self‑transcendence and lower on cooperativeness, suggesting either a protective effect of psychedelics or pre‑existing personality differences that predispose to regular use.

Hallucinogens in Mental Health: Preclinical and Clinical Studies on LSD, Psilocybin, MDMA, and Ketamine

This review summarises preclinical and clinical evidence that ketamine, psilocybin, LSD and MDMA have therapeutic potential for psychiatric disorders — with robust support for ketamine in depression, emerging evidence for psilocybin and LSD in treating depression and modulating functional brain connectivity, and MDMA showing efficacy in PTSD. It also delineates the pharmacology and behavioural effects of psychedelic versus non‑psychedelic hallucinogens and entactogens.
