Pharmacokinetics, Pharmacodynamics, and Urinary Recovery of Oral Mescaline Hydrochloride in Healthy Participants

This pharmacokinetic-pharmacodynamic analysis (n=46) of two Phase I trials of oral mescaline (100-800 mg) showed dose-proportional exposure with peak concentrations at 2 hours, a half-life of 3.5 hours, onset of effects around 1 hour post-dose, maximum effect intensity and duration ranging from 13% and 2.8 hours (100 mg) to 89% and 15 hours (800 mg), with 53% urinary excretion unchanged and 31% as the main metabolite, indicating at least 53% oral bioavailability.

Cross-sectional associations between lifetime use of psychedelic drugs and psychometric measures during the COVID-19 confinement: A transcultural study

In a cross‑sectional online survey of 2,974 English, Portuguese and Spanish speakers during the first COVID‑19 lockdown, regular lifetime users of psychedelic drugs reported lower psychological distress and peritraumatic stress and greater perceived social support than occasional users and non‑users. They also scored higher on novelty‑seeking and self‑transcendence and lower on cooperativeness, suggesting either a protective effect of psychedelics or pre‑existing personality differences that predispose to regular use.

Lifetime experience with (classic) psychedelics predicts pro-environmental behavior through an increase in nature relatedness

In a large-scale (N = 1,487) online study using structural equation modelling, lifetime experience with classic psychedelics uniquely predicted greater self-reported pro-environmental behaviour. This association was statistically mediated by increased nature relatedness and remained robust to controls for other drug use and personality traits, though the data are correlational.

The paradoxical psychological effects of lysergic acid diethylamide (LSD)

In a double-session, within-subjects study of 20 healthy volunteers, intravenous LSD (75 µg) produced acute psychosis‑like symptoms alongside heightened mood but led to increased optimism and trait openness two weeks later without raising delusional thinking. The authors propose that 5‑HT2A‑mediated increases in cognitive flexibility produce transient emotional lability during intoxication and leave a mid‑term “loosened cognition” that supports improved psychological wellbeing.
