Effects of hallucinogenic drugs on the human heart

This review summarises evidence that classic hallucinogens (bufotenin, psilocin/psilocybin, LSD, ergotamine/ergometrine, DMT and 5‑MeO‑DMT) act via cardiac 5‑HT receptors to produce positive inotropic and chronotropic effects in humans, increasing contractile force and heart rate and potentially provoking arrhythmias. It highlights these cardiovascular actions as an under‑appreciated risk of hallucinogen use.

Overcoming the clinical challenges of traditional ayahuasca: a first-in-human trial exploring novel routes of administration of N,N-Dimethyltryptamine and harmine

This open-label within-subject study (n=10) compared the oral administration of a capsule (containing DMT and harmine) with combined intranasal administration of an oromucosal harmine tablet and an intranasal DMT spray at two doses. The research aimed to improve the pharmacokinetics and tolerability profiles of ayahuasca-analogue formulations. Results indicate that the combined buccal/intranasal administration substantially attenuated common side effects and yielded significantly improved pharmacokinetic profiles. This suggests it may be an innovative approach for the safe and patient-oriented administration of DMT/harmine for treating affective disorders.

Pharmacological effects of methylone and MDMA in humans

In a randomised, double‑blind, placebo‑controlled crossover trial in 17 experienced psychostimulant users, a single oral dose of methylone (200 mg) produced sympathomimetic effects and stimulant/entactogenic subjective effects similar to MDMA (100 mg) but with a faster onset and shorter duration. These results indicate methylone’s acute pharmacological profile and abuse potential in humans are comparable to those of MDMA.

Changes in mental health, wellbeing and personality following ayahuasca consumption: Results of a naturalistic longitudinal study

In a naturalistic longitudinal study of 53 ayahuasca‑naïve attendees, a single facilitated ceremony produced broad 1‑month improvements in depression, anxiety, stress, substance use, personality traits, relationships and spirituality. Baseline characteristics—particularly high negative emotionality and body dissociation and low self‑efficacy—predicted greater benefit while facets of the mystical experience did not, indicating potential for personalised treatment and warranting randomised controlled trials.

Effects of 3,4-Methylenedioxymethamphetamine on Conditioned Fear Extinction and Retention in a Crossover Study in Healthy Subjects

In a double-blind, randomised, placebo-controlled crossover study in 30 healthy males, a single 125 mg dose of MDMA given after conditioning enhanced rapid fear extinction and retention as indexed by reduced skin conductance responses to the conditioned stimulus, but had no effect on fear‑potentiated startle. MDMA raised plasma oxytocin levels and produced subjective effects linked to reduced CS+/CS− discrimination, though oxytocin itself did not predict extinction outcomes, suggesting MDMA may facilitate certain forms of learned fear attenuation relevant to its therapeutic use.
