Adverse Events in Studies of Classic Psychedelics: A Systematic Review and Meta-Analysis

This systematic review and meta‑analysis of 214 studies (3,504 participants with analysable adverse‑event data) found that high‑dose classic psychedelics were generally well tolerated in clinical/research settings, with serious adverse events occurring mainly in ~4% of participants who had preexisting neuropsychiatric disorders and no reports in contemporary trials of suicide, persistent psychotic disorder or hallucinogen‑persisting perception disorder. Common non‑serious adverse events (headache, anxiety, nausea, fatigue, dizziness) had similar prevalences for psilocybin and LSD, but substantial heterogeneity and limited systematic adverse‑event monitoring across studies highlight the need for improved pharmacovigilance.

5-MeO-DMT for post-traumatic stress disorder: a real-world longitudinal case study

This first longitudinal case study reports that a single inhaled dose of toad‑derived 5‑MeO‑DMT produced rapid, clinically significant and sustained reductions in PTSD symptoms, hopelessness and suicide risk in a 23‑year‑old woman, with the participant endorsing a complete mystical experience and no drug‑related serious adverse events. Improvements persisted to 12 months but were accompanied by acute nausea, overwhelming subjective effects and later night terrors, and the authors emphasise the single‑case design is non‑generalizable and requires controlled replication.

Psychedelic Treatment for Trauma-Related Psychological and Cognitive Impairment Among US Special Operations Forces Veterans

In a retrospective survey of 51 U.S. Special Operations Forces veterans who completed a psychedelic programme in Mexico (ibogaine and 5‑MeO‑DMT), participants reported very large reductions in suicidal ideation, PTSD, depression, anxiety and cognitive impairment alongside large increases in psychological flexibility. These preliminary, self‑reported findings are limited by retrospective design and lack of randomisation/blinding, so controlled trials are needed to establish efficacy.
